ABSTRACT

Objective To analyze the correlation between electrocardiogram exercise stress test and coronary artery stenosis degree.Methods A total of 150 patients who performed electrocardiogram exercise stress test were selected,and then coronary angiography were performed within 2 weeks.The sensitivity,specificity,positive predictive value and accuracy of electrocardiogram exercise stress test for diagnosing coronary artery stenosis degree were analyzed.Results Compared with coronary angiography,the sensitivity,specificity,positive predictive value and accuracy of electrocardiogram exercise stress test for diagnosing coronary heart disease were 75%(48/64),74% (64/86),69%(48/70) and 75% (112/150),respectively,for diagnosing coronary artery disease were 31% (19/62),42% (37/88),27% (19/70) and 37% (56/150),respectively.Conclusions Electrocardiogram exercise stress test as a simple,feasible and noninvasive method,has a good concordance with coronary angiography and a high correlation with coronary heart disease,a low correlation with coronary artery disease.It can be used as a good way to screen patients with coronary heart disease.

ABSTRACT

Objective:To explore the influence of chronic cobalt chloride (CoCl2 ) treatment on blood glucose and car-diomyocyte apoptosis in diabetic rats .Methods:In vivo MI model of rat was established using male GK diabetic rats and male Wistar rats (all weighing 200g~250g) .Blood glucose level was measured before model establishment .Af-ter successful operation ,rats were divided into six groups:Wistar sham operation group ,Wistar MI group ,Wistar MI + CoCl2 group ,GK sham operation group ,GK MI group and GK MI + CoCl2 group ,six rats survived in each group at least .After three weeks ,blood glucose level was measured and heart was excised ,expression of Caspase-3 apoptosis gene in ischemic necrosis area was measured .Results:(1) Compared with GK sham operation group and GK MI group on three weeks after operation ,there was significant reduction in blood glucose level [ (27.73 ± 2.58) mmol/L ,(27.87 ± 3.18) mmol/L vs .(16.3 ± 2.15) mmol/L] in GK MI + CoCl2 group ,P0.05 all .Conclusion:Chronic cobalt chloride treatment can decrease blood glucose level and reduce expression of Caspase-3 apoptosis gene in ischemic necrosis area in diabetic rats .

ABSTRACT

Objective:To explore whether ginsenosides-Rb1 (Gs-Rb1)can relieve cardiomyocyte hypertrophy induced by endothelin-1 (ET-1)via protein kinase C (PKC)system.Methods:Cardiomyocytes of neonatal rat were random-ly divided into blank control group,Gs-Rb1 group,ET-1 group,Gs-Rb1+ET-1 group,ET-1+CHE (chelerythrine, PKC blocker)group and Gs-Rb1 +ET-1 +CHE group.After 96h intervention,cardiomyocyte surface area,total protein content,PKC activity,c-fos and p-c-jun expressions were measured.Results: (1)Cardiomyocyte surface area and total protein content in Gs-Rb1+ET-1 group were significantly lower than those of ET-1 group (P <0.05~<0.001),but not significant different with those of Gs-Rb1+ET-1+CHE group,P =0.569;(2)PKC activity in Gs-Rb1+ET-1 group was significantly lower than that of ET-1 group [(9.3±0.6)pmol·min-1 ·mg-1 vs.(14.1± 0.9)pmol·min-1 ·mg-1 ],but significantly higher than that of Gs-Rb1+ET-1+CHE group [(2.7±0.2)pmol· min-1 ·mg-1 ],P <0.001 all;(3)Expressions of c-fos and p-c-jun gene and protein in ET-1 group were significant-ly higher than those of blank control group (P <0.001 all);compared with ET-1 group,there were significant re-ductions in expressions of c-fos [mRNA/protein:(0.53±0.05/0.39±0.02)vs.(0.43±0.03/0.31±0.03)]and p-c-jun [mRNA/protein:(0.64±0.04/0.44±0.02)vs.(0.33±0.05/0.37±0.03)]in Gs-Rb1+ET-1 group and ex-pressions of c-fos [mRNA/protein:0.41 ± 0.05/0.31 ± 0.02]and p-c-jun [mRNA/protein:0.31 ± 0.05/0.36 ±0.03]in ET-1+CHE group (P <0.05 or <0.001),expressions of c-fos and p-c-jun gene and protein in Gs-Rb1+ET-1+CHE group were significantly lower than those of Gs-Rb1+ET-1 group and ET-1+CHE group (P <0.05 or<0.001).Conclusion:Gs-Rb1 can significantly inhibit cardiomyocyte hypertrophy induced by ET-1 and PKC system is one of pathways mediating this biological effect.

ABSTRACT

BACKGROUND:Current cardiac bioprostheses and mechanical valves are shown to have some flaws and shortcomings, and tissue-engineered heart valves which can avoid these problems are becoming an ideal choice for valve replacement. OBJECTIVE:To explore the experimental progress in the construction of tissue-engineered heart valves. METHODS:Experimental studies related to tissue-engineered heart valves were retrieved in databases. Three main elements for tissue-engineered heart valves are seed cells, scaffold materials, and cellseeding. RESULTS AND CONCLUSION:Cardiac valve repair and replacement is the primarily surgical treatment for valvular heart disease. At present, seed cells mainly for construction of tissue-engineered heart valves include vascular endothelial cells, endothelial progenitor cells and bone marrow mesenchymal stem cells. The acellular scaffold has good biomechanical properties and histocompatibility. After cellseeding, a continuous celllayer wil form on the scaffold surface, which makes it possible to construct tissue-engineered heart valves. Tissue-engineered heart valve has a good prospect, but it is stil in the early stage of research and there are stil many problems that need to be solved.

ABSTRACT

BACKGROUND: The negative emotion can directly attenuate the humoral and cell mediated immunity, which contributing to the onset of lung cancer,therefore psychological therapy is crucial for the prevention of lung cancer.OBJECTIVE: To investigate the effect of catharsis and cognitive therapy on the psychology of patients with lung cancer, as well as the short-term clinical therapeutic effect, so as to exploring the importance of psychological interventionDESIGN: Non-randomized same period control, 1:1 matched observation SETTING: Cadres Therapeutic Ward of Affiliated Second Hospital of China Medical University and Cadres Therapeutic Ward of General Hospital of Shenyang Military Area Command of Chinese PLA.PARTICIPANTS: Totally 124 patients with lung cancer, received treatment at Cadres Therapeutic Ward of Affiliated Second Hospital in China Medical University and General Hospital of Shenyang Military Area Commaud of Chinese PLA from February 1999 to December 2002, volunteered into this study, were randomly divided into treatment group and control group, all data was paired by 1:1 according to age, gender, UICC-TUM stage and pathological type, totally 42 pair data were obtained.METHODS: All participant received the same chemotherapy and radiotherapy (etoposide carboplatin), the patients in treatment group were given additional emotional catharsis and cognitive therapy at the beginning of treatment once every five days for 1-3 times; And then patients were informed of related cancer knowledge once or twice a week for totally 6 weeks. Before treatment, SCL-90 (including 9 symptomatic factors that divide into five grades: 0 score meant never, 1 score was gentle, 2 scores were medium, 3 scores were quite severe and 4 scores were severer) was used to evaluate the psychology of patients; moreover the therapeutic effect was assessed according to WHO standards for lung cancer (focus disappeared for more than 4 weeks is believed of complete relieve; Focus area reduced by above 50% and maintained for 4 weeks is believed of part relieve) MAIN OUTCOME MEASURES: Scores for SCL-90 and therapeutic effect of two groups before and after treatment RESULTS: Totally 42 pairs (84 cases) were remained in the final result duced after treatment in two groups (P < 0.05), but the scores for somatzation, individual sensitivity, depression anxiety and psychosis in treatment group were found lower than corresponding control group after treatment (1.64±0.41, 1.76±0.34; 1.31±0.30, 1.67±0.35; 1.40±0.29, 1.58±0.41;Therapeutic effect: The total relieve rate in treatment group was higher than control group, but difference was of no statistical significance (68%,52%, x2=2.381, P > 0.05).CONCLUSION: Although the therapeutic effect of catharsis and cognitive therapy is s unobvious short-term in patients with lung cancer, but it can attenuate the negative emotion, such as depression and anxiety, which can improve their psychology and immune-regulating function.

ABSTRACT

<p><b>OBJECTIVE</b>To identify and characterize laryngeal cancer related novel genes located on chromosome 6q25.</p><p><b>METHODS</b>Electric hybridization was performed in human genome database using EST (expression sequence tag) as probe. Novel genes were deduced by software from positive DNA clones and their cDNAs were amplified by RT-PCR using primers designed according to the sequence of the putative genes.</p><p><b>RESULTS</b>A novel gene was cloned successfully. The full length of this gene was about 21 kb. It contained two exons and produced a 1006 bp transcript coding a protein with 235 amino acid residues. It's 5'flanking sequence contained two binding sites of oncoprotein c-Myc, thus it was named MTLC (c-Myc target from laryngeal cancer cells). Homologous assay showed that MTLC exhibited little overall homology to known human proteins but it exhibited good overall homology to mouse MT-MC1 protein with an identity of 78%. The primary structure of MTLC protein contained a nuclear location signal motif, but it did not have other conserved domains. The results of subcellular location experiment showed that MTLC expressed in nuclei of human hepatocellular carcinoma cell line Bel7402 cells, while a wide distribution of MTLC in various tissues was demonstrated by Northern blotting.</p><p><b>CONCLUSION</b>MTLC may play an important role as a target gene of c-Myc and as a transcription factor in keeping the normal physiological process of cells.</p>
